چکیده

Abstract Multiple morphological abnormalities of the sperm flagella (MMAF) is one major causes male infertility and characterized by multiple defects. In this study, we found that coiled-coil domain-containing 189 (Ccdc189) gene was predominantly expressed in mouse testes inactivation Ccdc189 caused infertility. Histological studies revealed most from Ccdc189-deficient mice carried coiled, curved, or short flagella, which are typical MMAF phenotypes. Immunoelectron microscopy showed CCDC189 protein located at radial spoke first peripheral microtubule doublets (MTDs) axoneme. A CCDC189-interacting protein, CABCOCO1, discovered via coimmunoprecipitation mass spectrometry, Cabcoco1 malformation consistent with findings obtained mice. Further downregulation CABCOCO1 expression both interacted spoke-specific RSPH1 intraflagellar transport proteins. This study demonstrated a spoke-associated involved flagellum formation through its interactions

Iqcg Is Essential for Sperm Flagellum Formation in Mice

Mammalian spermatogenesis comprises three successive phases: mitosis phase, meiosis phase, and spermiogenesis. During spermiogenesis, round spermatid undergoes dramatic morphogenesis to give rise to mature spermatozoon, including the condensation and elongation of nucleus, development of acrosome, formation of flagellum, and removal of excessive cytoplasm. Although these transformations are wel...

I-3: Tale of The Tail: Candidate Genes Involved in Sperm Flagella Formation

Background ISTS defect in which sperm tail is short and fibrous sheath and axoneme are disorganized, is one of the syndromes that cause male infertility. Although a few studies have been done in this regard, its exact etiology in human is unclear yet. Four candidate genes causing ISTS are SPEF2, RABL2B, and A-kinas anchoring proteins genes (AKAP3 and AKAP4). Proteins which coded by SPEF2 and RA...

A MAP6-Related Protein Is Present in Protozoa and Is Involved in Flagellum Motility

In vertebrates the microtubule-associated proteins MAP6 and MAP6d1 stabilize cold-resistant microtubules. Cilia and flagella have cold-stable microtubules but MAP6 proteins have not been identified in these organelles. Here, we describe TbSAXO as the first MAP6-related protein to be identified in a protozoan, Trypanosoma brucei. Using a heterologous expression system, we show that TbSAXO is a m...

Basal Body Positioning Is Controlled by Flagellum Formation in Trypanosoma brucei

To perform their multiple functions, cilia and flagella are precisely positioned at the cell surface by mechanisms that remain poorly understood. The protist Trypanosoma brucei possesses a single flagellum that adheres to the cell body where a specific cytoskeletal structure is localised, the flagellum attachment zone (FAZ). Trypanosomes build a new flagellum whose distal tip is connected to th...
